人BMP-7基因定位于2號(hào)染色體,其cDNA編碼區(qū)有1293個(gè)堿基,含有1個(gè)開放讀碼框,編碼多肽產(chǎn)物由431個(gè)氨基酸殘基組成,起始序列為一段由29個(gè)氨基酸殘基組成信號(hào)肽,中間第29-292個(gè)氨基酸殘基稱為前肽,第293-431個(gè)氨基酸殘基稱為成熟肽。該前體蛋白合成分泌后,在體內(nèi)被剪切加工成漢139個(gè)氨基酸殘基成熟肽,每個(gè)成熟有活性BMP-7都是由二硫鍵連接成同源或異源二聚體。BMP-7具有多方面生物學(xué)效應(yīng),包括在胚胎發(fā)生3個(gè)階段發(fā)揮作用,在腎臟形態(tài)發(fā)生中也起到重要作用,另外BMP-7還具有誘導(dǎo)成骨作用,能促成骨細(xì)胞增殖分化,可誘導(dǎo)軟骨細(xì)胞分化,在骨代謝中發(fā)揮重要作用。
人骨形態(tài)發(fā)生蛋白-7,CHO表達(dá)重組蛋白是在CHO中國(guó)倉(cāng)鼠卵巢細(xì)胞中重組表達(dá)的。其序列結(jié)構(gòu)為“N末端--人BMP-2蛋白(Met 1- Arg 282)--人BMP-7蛋白(Ser 293 - Arg 431)--C末端”。表達(dá)人BMP-2的信號(hào)肽和1-282個(gè)氨基酸序列與人BMP-7成熟肽293-431個(gè)氨基酸序列融合表達(dá)于CHO細(xì)胞。最后成熟的BMP-7蛋白是通過(guò)蛋白酶水解掉包含信號(hào)肽和前肽的139個(gè)氨基酸而形成。糖基化增大了人骨形態(tài)發(fā)生蛋白-7分子量,使非還原狀態(tài)下在SDS-PAGE電泳時(shí)條帶位于25-40kDa。該人骨形態(tài)發(fā)生蛋白-7,CHO表達(dá)重組蛋白最終由標(biāo)準(zhǔn)的色譜技術(shù)純化出。

溶解:提供的人骨形態(tài)發(fā)生蛋白-7,CHO表達(dá)(BMP 7 CHO)產(chǎn)品為凍干粉末;該凍干粉末是從含1mg/ml人骨形態(tài)發(fā)生蛋白-7,CHO表達(dá)(BMP 7 CHO)的1%蔗糖,1.2%甘露醇,20mM 氨基乙酸和0.05% Tween 20 (pH 4)溶液中滅菌并凍干產(chǎn)生的。我們建議加1ml無(wú)菌且不含內(nèi)毒素的水以配制工作儲(chǔ)存液。
保存:人骨形態(tài)發(fā)生蛋白-7,CHO表達(dá)(BMP 7 CHO)凍干粉末在室溫可保存3周,或置于-18℃凍存用于長(zhǎng)期保存。重新溶解后可以在4℃冷藏保存至少2-7天以上;如果想延長(zhǎng)保存時(shí)間,建議分裝(加載體蛋白如0.1% HSA或 BSA)后置于-18℃凍存。請(qǐng)避免反復(fù)凍融。

Introduction (介紹)
The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development. In addition, the fact that this BMP is closely related to BMP5 and BMP7 has lead to speculation of possible bone inductive activity.
Description (描述)
N-TERMINAL---Human BMP-2 (Met 1 – Arg 282) Human BMP-7 (Ser 293 – Arg 431)---C-TERMINAL. The DNA sequence encoding the human BMP-2 signal peptide and propeptide (1~282 amino acid) fused to the human rhBMP-7 mature chain (293~431 amino acid) was expressed in a Chinese hamster ovary cell line. The mature recombinant BMP-7 generated by the proteolytic removal of the signal peptide and propetide contains 139 amino acid residues. The glycosylation of BMP-7 increases the molecular mass and the glycosylated proteins migrate as 25 ~ 40 kDa in SDS-PAGE under non-reducing conditions.
BMP-7 is purified by proprietary chromatographic techniques.

Formulation
BMP-7 was lyophilized from a concentrated (1mg/ml) sterile solution containing 1% sucrose, 1.2% mannitol, 20mM glycine, and 0.05% tween 20 pH-4.
Solubility (溶解性)
It is recommended to reconstitute the lyophilized Bone Morphogenetic Protein-7 in 1ml sterile/ endotoxin free water.
Stability (穩(wěn)定性)
Lyophilized BMP-7 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution BMP 7 Human should be stored at 4°C between 2-7 days and for future use below -18°C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.
Purity (純度)
Greater than 97.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.
Biological Activity (生物活性)
Measured in alkaline phosphatase activity assay using MC3T3-E1 cells. The ED50 for this effect is <100ng/ml, corresponding to a Specific Activity of 10,000IU/mg.
