肝細(xì)胞生長因子（hepatocyte growth factor，HGF）最初作為一種肝細(xì)胞有絲分裂原是從肝部分切除大鼠的血清中分離得到的。HGF是來源于間充質(zhì)的多效生長因子，可以促進(jìn)多種細(xì)胞的分裂和生長。以旁分泌、內(nèi)分泌、自分泌形式在保護(hù)器官，延長組織老化方面發(fā)揮作用。HGF首先以無活性的單鏈蛋白（HGF SF）形式分泌，經(jīng)肝細(xì)胞生長因子激活因子切割成成熟的異二聚體HGF，機(jī)體內(nèi)HGFA可激活HGF SF前體，使之從單鏈前體變成雙鏈活性形式，是調(diào)節(jié)細(xì)胞外HGF SF活性的關(guān)鍵步驟。HGF在促進(jìn)肝細(xì)胞再生的同時(shí)參與正常組織的損傷后修復(fù)、細(xì)胞的運(yùn)動(dòng)以及腫瘤的形成、浸潤和轉(zhuǎn)移、分化以及腫瘤血管行程中發(fā)揮重要的作用。

溶解：提供的人肝細(xì)胞生長因子,CHO表達(dá)(HGF CHO)產(chǎn)品為過濾滅菌的白色凍干粉末；該凍干粉末是從含1mg/ml人肝細(xì)胞生長因子,CHO表達(dá)(HGF CHO)的0.02% Tween 80，磷酸鹽緩沖液（pH 7.4）中凍干產(chǎn)生的。我們建議加無菌的18MΩ-cm水以配制工作儲(chǔ)存液到100ug/ml，后繼實(shí)驗(yàn)過程中可根據(jù)實(shí)驗(yàn)要求進(jìn)一步稀釋。
保存：人肝細(xì)胞生長因子,CHO表達(dá)(HGF CHO)凍干粉末在室溫可保存3周，或置于-18℃凍存用于長期保存。重新溶解后可以在4℃冷藏保存至少2-7天；如果想延長保存時(shí)間，建議分裝（加載體蛋白如0.1% HSA或 BSA）后置于-18℃凍存。請避免反復(fù)凍融。

Introduction 

Hepatocyte Growth Factor (HGF) is a multifunctional growth factor which regulates both cell growth and cell motility. It exerts a strong mitogenic effect on hepatocytes and primary epithelial cells. HGF synergizes with Interleukin-3and GM-CSFto stimulate colony formation of hematopoietic progenitor cells in vitro and may, therefore, also modulate hematopoiesis.

Description 

Hepatocyte Growth Factor Human Recombinant produced in CHO is a heterodimer, non-glycosylated, polypeptide chain consisting an a-chain of 463 amino acids and b-chain of 234 having a total molecular mass of 75kDa.
The HGF is purified by proprietary chromatographic techniques.

Formulation 

The protein was lyophilized from a concentrated solution containing Phosphate-Buffered Saline with 0.02% Tween 80, pH 7.4.

Solubility  （溶解性）

It is recommended to reconstitute the lyophilized Hepatocyte Growth Factor in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability    （穩(wěn)定性）

Lyophilized Hepatocyte Growth Factor although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution HGF should be stored at 4°C between 2-7 days and for future use below -18°C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

Greater than 97.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.

Biological Activity    （生物活性）

The ED50, calculated by the dose-dependant proliferation of monkey 4MBr-5 indicator cells was found to be 20-40ng/ml corresponding to a Specific Activity of 25,000-50,000IU/mg.

Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 1.83 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of HGF as a Reference Standard.
