血小板衍生生長因子（Platelet Derived Growth Factor，PDGF）是由兩條相關(guān)多肽鏈通過二硫鍵連接而成的二聚體。PDGF 貯存于血小板α顆粒中的一種堿性蛋白質(zhì)，是一些細胞，如纖維母細胞、平滑肌細胞、神經(jīng)膠質(zhì)細胞的促細胞分裂劑。PDGF可分為PDGF-I和PDGF-II，二者所含蛋白質(zhì)相同，僅糖苷含量各異，它們是由兩個不同的多肽鏈由二硫鍵結(jié)合而成的二聚體，其中A鏈較大(PDGF-I中的A鏈為17 000，PDGF-II中的A鏈為15 000)，B鏈較小(14 000)。由血小板純化得到的PDGF為AB、BB雙聚體，許多腫瘤細胞中的PDGF為AA雙聚體。目前認為PDGF在傷口愈合、膽固醇與磷酯合成的調(diào)節(jié)、動脈粥樣硬化等多種生理及病理過程中起重要作用。

溶解：提供的人血小板衍生生長因子-BB,酵母表達(PDGF BB Yeast)產(chǎn)品為經(jīng)0.2um孔徑過濾滅菌的白色凍干粉末；該凍干粉末是從含人血小板衍生生長因子-BB,酵母表達(PDGF BB Yeast)的20mM 乙酸鈉（不含任何載體蛋白）溶液中過濾滅菌并凍干產(chǎn)生的。我們建議加無菌的18MΩ-cm水以配制工作儲存液到不低于100ug/ml，后繼實驗過程中可根據(jù)實驗要求進一步稀釋。
保存：人血小板衍生生長因子-BB,酵母表達(PDGF BB Yeast)在室溫可保存3周，或置于-18℃凍存用于長期保存。重新溶解后可以在4℃冷藏保存至少2-7天；如果想延長保存時間，建議分裝后置于-18℃凍存。請避免反復凍融。

Introduction  （介紹）

PDGF-BB is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a motif of eight cysteines. This gene product can exist either as a homodimer (PDGF-BB) or as a heterodimer with the platelet-derived growth factor alpha polypeptide (PDGF-AB), where the dimers are connected by disulfide bonds. Mutations in this gene are associated with meningioma. Reciprocal translocations between chromosomes 22 and 7, at sites where this gene and that for COL1A1 are located, are associated with a particular type of skin tumor called dermatofibrosarcoma protuberans resulting from unregulated expression of growth factor. Two splice variants have been identified for this gene.

Description  

Platelet-Derived Growth Factor BB Human Recombinant is a glycosylated homodimer produced in Saccharomyces cerevisiae, containing 2x109 amino acids and having a molecular mass of 32kDa.
PDGF-BB is purified by proprietary chromatographic techniques.

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation 

The PDGF-BB was lyophilized from 0.2 µm filtered solution in 20mM sodium phosphate in absence of any carrier protein.

Solubility  （溶解性）

It is recommended to reconstitute the lyophilized Platelet-derived Growth Factor-BB in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability   （穩(wěn)定性）

Lyophilized Platelet-derived Growth Factor BB although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution PDGF BB should be stored at 4°C between 2-7 days and for future use below -18°C.

Greater than 98.0% as determined by SDS-PAGE.

Amino acid sequence   （氨基酸序列）

The sequence of the first five N-terminal amino acids was determined and was found to be Ser-Leu-Gly-Ser-Leu.

Biological Activity   （生物活性）

The ED50, as measured by its ability to stimulate proliferation of mouse 3T3 fibroblasts is typically 1 ng/ml, corresponding to a Specific Activity of 1,000,000IU/mg.
