MycoAlert? PLUS Assay是一種精選生物化學(xué)檢測(cè)方法，用于檢測(cè)細(xì)胞培養(yǎng)過(guò)程中的支原體污染問(wèn)題。支原體是一種能夠自我復(fù)制的最小的原核微生物，和其他細(xì)菌不同的是它們沒(méi)有細(xì)胞壁，所以細(xì)胞形態(tài)不固定，可以有多種變化，可以在培養(yǎng)基上形成極小的菌落。支原體是寄生生物，但同時(shí)也具有腐生功能。細(xì)胞培養(yǎng)過(guò)程中的污染支原體會(huì)使細(xì)胞生長(zhǎng)變慢，影響細(xì)胞活性變化。新一代MycoAlert? PLUS Assay可以檢測(cè)出所有6種支原體屬，多達(dá)180種支原體。與第一代MycoAlert?支原體檢測(cè)試劑盒相比有更高的靈敏度和準(zhǔn)確性。檢測(cè)樣品中的活性支原體裂解后，釋放出的酶與MycoAlert? PLUS Substrate反應(yīng)，催化ADP轉(zhuǎn)化為ATP。加入MycoAlert? PLUS　Reagent試劑后在熒光素酶作用下ATP產(chǎn)生光信號(hào)(ATP+ Luciferin + O2 →Oxyluciferin + AMP + PPi + CO2 + LIGHT)。光信號(hào)可用光度計(jì)讀出，根據(jù)第二次讀數(shù)(Read B)與第一次讀數(shù)(Read A)比率可是確定是否存在支原體污染。Read B/Read A比率<1, 支原體陰性，無(wú)支原體；1-1.2，不確定是否存在支原體，需重新檢測(cè)；>1.2，支原體陽(yáng)性，存在支原體污染。

MycoAlert? PLUS Mycoplasma Detection Kit:
Mycoplasma are the smallest and simplest prokaryotes. Mycoplasma depend on their hosts for
many nutrients due to their limited biosynthetic capabilities. They have long been recognized as
common contaminants of cells in continuous culture, but their presence may go undetected for months. As the mycoplasma competes with the cells for the nutrients in culture media, one of the first signs is a reduction in the rate of cell proliferation and slight changes in cellular responses, including gene expression.
Mycoplasma detection in cell cultures has, until now, been a long, drawn out process with difficult-tointerpret results. The MycoAlert? PLUS Kit is intended for the quick and convenient detection of viable mycoplasma in cell cultures. The speed and convenience of the MycoAlert? PLUS Kit allows for the routine testing of cells in culture and commonly used constituents of complete media.
To allow for the early detection of mycoplasma contamination, Lonza recommends testing at every cell passage. Frequent testing such as this will indicate when a cell line becomes infected, allowing prompt remedial action to be taken. The MycoAlert? PLUS Assay can also be extended to incoming cell lines and the commonly used constituents of complete media.
