LONZA鱟試劑產品－Gel Clot LAL Assay產品介紹

鱟阿米巴樣細胞裂解物（ Limulus Amebocyte Lysate,LAL ）被用于檢測革蘭氏陰性細菌細胞壁的成分脂多糖（內毒素）。該裂解物來源于鱟（Limulus polyphemus）的循環(huán)阿米巴樣細胞。

使用 LAL 進行內毒素檢測來源于 Bang 的發(fā)現， Bang 觀察發(fā)現 Limulus polyphemus 革蘭氏陰性感染后導致致命性的血管內凝聚。 Levin 和 Bang 后來發(fā)現這種凝結現象是內毒素與鱟血中的循環(huán)阿米巴樣細胞中的一種可凝結蛋白相互作用的結果。 Levin 和 Bang 從洗滌后的阿米巴樣細胞中制備了一種裂解物，該裂解物是內毒素的一種非常敏感的指示劑。 Solum,Young,Levin 和 Prendergast 已經從 LAL 中純化和定性了可凝結蛋白，并且可與內毒素發(fā)生酶促反應。

第一個商業(yè)化 LAL 檢測分析是凝膠分析。 1977 年， FDA （生物學）確定了一個裂解物的參考標準用于確定每批商業(yè)化裂解物的敏感度。他們隨后出版了詳細的實驗室操作流程用于每批裂解物和標準品的比較分析。
當前有四種LAL法已經獲得 FDA 的批準。第一種，凝膠法，LONZA的商標名稱為 PYROGENT,是基于內毒素存在的條件下發(fā)生LAL凝結現象的事實。動力學濁度法，LONZA商標名稱為 PYROGENT-5000 ，是一種利用凝膠率來測定內毒素含量的定量LAL法。第三種和第四種方法即顯色法， 美國LONZA公司商標名稱為 QCL-1000 和 Kinetic-QCL ，采用一種在有 LAL 和內毒素的情況下能顯黃色的顯色底物來定量 LAL ，顏色的深淺與內毒素的濃度呈線性相關。
